Drosophila embryo collection
1)For each genotype that you want to collect prepare 1 apple juice plate and one cup.
a.Warm the apple juice plate to room temperature (RT) and put a dollop (about dime size) of yeast paste on the middle of the plate.
b.Flies like fresh yeast paste. Be sure that the yeast is less than 2 weeks old.
c.Be sure the cup(s) you picked out have air holes in the top.
2)Turn the cup upside down and insert a funnel. Knock the flies down to the bottom of their bottle or vial, flip the bottle or vial upside down into the funnel, then quickly tap the flies down into the cup, remove the funnel and replace it with a plate.
a.If putting multiple vials worth of flies into the same cup, keep tapping the flies in the cup down between vials.
b.Put a piece of tape on the bottom of the plate for a label and to allow you to pull the plate off later.
c.Flies will not lay well the first day they are in a cup. Don’t panic, just put a fresh plate on and wait for the next day.
